Light exposure of the ovum and preimplantation embryo during ART procedures [2007 archive]
95% of the radiation was from microscopes. Ambient light, in contrast, was not a significant contributor to light stress and the use of dark laboratories is not justified.
Comprehensive chromosome screening with synchronous blastocyst transfer: time for a paradigm shift
Perhaps most prominent has been the resurgence of embryonic aneuploidy screening. The validation of 24-chromosome testing platforms has led to a variety of studies demonstrating higher implantation and delivery rates. These findings are now translating to changes in the paradigm of ART practice.
Pesticide Methoxychlor Promotes the Epigenetic Transgenerational Inheritance of Adult-Onset Disease through the Female Germline
Analysis of the F3 generation sperm epigenome of the methoxychlor lineage males identified differentially DNA methylated regions (DMR) termed epimutations in a genome-wide gene promoters analysis. These epimutations were found to be methoxychlor exposure specific in comparison with other exposure specific sperm epimutation signatures. Observations indicate that the pesticide methoxychlor has the potential to promote the epigenetic transgeneration
Appropriate Crypt Formation in the Uterus for Embryo Homing and Implantation Requires Wnt5a-ROR Signaling
Embryo homing and implantation occur within a crypt (implantation chamber) at the antimesometrial (AM) pole along the uterus. The mechanism by which this is achieved is not known. Here, we show that villi-like epithelial projections from the main uterine lumen toward the AM pole at regularly spaced intervals that form crypts for embryo implantation were disrupted in mice with uterine loss or gain of function of Wnt5a, or loss of function of both
Effect of micro-vibration culture system on embryo development
Micro-vibration culture methods have a beneficial effect on embryonic development in mouse embryos. In poor (human) responders, the embryo development rate was improved to a limited extent under the micro-vibration culture conditions, but the clinical results were significantly improved.
In-vitro culture of human embryos with mechanical micro-vibration increases implantation rates [archive 2011]
This review covers the history of this question and in this light offers an explanation through biological concept for one of the most recent developments in this area: in-vitro culture of human embryos with mechanical micro-vibration. The effect of mechanical micro-vibration on embryos during their in-vitro culture was examined. Pregnancy rates after the transfer of embryos in the group with in-vitro culture under mechanical vibration were incr
Human oocyte-derived sperm chemoattractant is a hydrophobic molecule associated with a carrier protein
Oocyte-conditioned media treated with proteases did not lose their chemotactic activity: on the contrary, they became more active, with the activity shifted to lower concentrations. When oocyte-conditioned media were subjected to hexane extraction, chemotactic activity was found in both the hydrophobic and aqueous phases. Known mammalian sperm chemoattractants were ruled out as oocyte-derived chemoattractants.
Perinatal outcomes in 6,338 singletons born after intrauterine insemination in Denmark, 2007 to 2012: the influence of ovarian stimulation
Singletons born after IUI had higher risk of adverse perinatal outcomes compared with SC children, similar to ICSI, but favorable outcomes compared with IVF. Stimulation with clomiphene citrate was associated with higher risk of SGA compared with natural-cycle IUI, but follicle-stimulating hormone treatment did not seem to be associated with adverse outcomes.
Physical activity is not related to semen quality in young healthy men
In contrast to previous research among athletes, these data suggest that physical activity is not deleterious to testicular function, as captured by semen quality parameters in this population of healthy young men in Spain.
Examining the temperature of embryo culture in in vitro fertilization: a randomized controlled trial comparing traditional core temperature (37°C) to
A total of 805 mature oocytes were cultured: 399 at 36°C and 406 at 37°C. Paired analysis demonstrated a higher rate of usable blastocyst formation per zygote at 37°C (48.4%) vs. at 36°C (41.2%). Rates of fertilization, aneuploidy, and sustained implantation were equivalent.
Effect of local endometrial injury on pregnancy outcomes in ovum donation cycles
his is the first study done in ovum recipients who underwent LEI by a “scratch” procedure after failed implantation. Unlike most previous reports, which found improved pregnancy rates with the use of “scratch effect” or “minor endometrial injury” after repeated implantation failures in standard IVF with own eggs, we did not find any changes in implantation rates in a population of egg recipients following this procedure. In view of a possible pos
Obstetric and perinatal outcome of babies born from vitrified oocytes
Conclusion(s): Although our data, the largest series to date, suggest that oocyte vitrification does not increase adverse obstetric and perinatal outcomes in children conceived with vitrified oocytes, further studies with larger samples are required to reinforce our conclusions.
Pesticide linked to three generations of disease: Methoxychlor causes epigenetic changes
The work is also the first to show that a majority of transgenerational disease traits can be transmitted primarily through the female line.
Additionally, the study identified mutations in the sperm epigenome of great-grandchild male rats. The epigenome functions like a set of switches for regulating gene expression and can be altered by environmental conditions.
Molecular key to healthy pregnancy discovered by researchers
A crucial molecular key to healthy embryo implantation and pregnancy has been discovered in a study that may offer new clues about the medical challenges of infertility/subfertility, abnormal placentation, and placenta previa. The authors found that uterine expression of a gene called Wnt5a - a major signaling molecule in cell growth and movement in both embryo development and disease - is critical to healthy embryo implantation in the uterus.
With China's relaxation of its one-child policy, more than 20 couples are now applying to have their second test-tube babies using their frozen embryos, one couple of which have stored their embryos for up to six years.
DIY Vaginal Ultrasounds Could Reduce Trips to the Doctor
A new "do-it-yourself" ultrasound method could let women who are undergoing in vitro fertilization (IVF) cut down on their trips to the doctor's office, a new study suggests.

A randomized controlled study of human serum albumin and serum substitute supplement as protein supplements for IVF culture and the effect on live birth rates. HR (2009)
Comment
I came across this reference in HR after reading the chapter on Macromolecules in EMBRYO CULTURE by Smith et.al. This chapter and the above article highlight a continuing problem in IVF media in that there remains considerable variation in the macromolecules used in IVF. This will become more important when using time-lapse imagery to identify the best embryos. It would seem to me that the impacts of media formulations on cleavage rates need to be resolved before commercial time points are set. Having said this, most time points are for the early cleavage benchmarks where the influence of macromolecules may be of less importance.
