Sperm DNA damage and assisted reproductive technologies: reasons to be cautious!
The objective of ART should not be simply taking a baby home. It should go beyond that endpoint and the target should be taking home a healthy baby who will become a healthy child and a healthy adult. Knowing that sperm DNA alterations may challenge these issues and that ART itself may induce sperm DNA alterations [3], particular caution should be taken in using and selecting spermatozoa that show the least possible level of DNA alterations.
Predictive value of sperm deoxyribonucleic acid (DNA) fragmentation index in male infertility
Conclusion: There was significant difference between the median value of sperm DFI of infertile men and fertile men. Compared to semen analysis, sperm DFI at cut-off point of 26.1% has a higher diagnostic value (AUC).
`This is where it all started` – the pivotal role of PLCζ:: within the sophisticated process of mammalian reproduction: a systemic review
Fertilization failure is attributed to lack of oocyte second meiosis resumption, suggesting that ICSI failure may be related to impaired PLCζ:: activity. Microinjection of recombinant human PLCζ:: to human oocytes after ICSI fertilization failure may trigger Ca2+ oscillations and achieve successful fertilization, offering new hope for couples traditionally referred to sperm donation.
Healthy offspring from freeze-dried mouse spermatozoa held on the International Space Station for 9 months
Radiation on the International Space Station (ISS) is more than 100 times stronger than at the Earth’s surface, and at levels that can cause DNA damage in somatic cell nuclei. The damage to offspring caused by this irradiation in germ cells has not been examined, however. Here we preserved mouse spermatozoa on the ISS for 9 mo. Although sperm DNA was slightly damaged during space preservation, it could be repaired by the oocyte cytoplasm and did
A bioprosthetic ovary created using 3D printed microporous scaffolds restores ovarian function in sterilized mice
Follicle-seeded scaffolds become highly vascularized and ovarian function is fully restored when implanted in surgically sterilized mice. Moreover, pups are born through natural mating and thrive through maternal lactation. These findings present an in vivo functional ovarian implant designed with 3D printing, and indicate that scaffold pore architecture is a critical variable in additively manufactured scaffold design for functional tissue engi
A microfluidic culture model of the human reproductive tract and 28-day menstrual cycle
The reproductive tract tissues and peripheral organs integrated into a microfluidic platform, termed EVATAR, represents a powerful new in vitro tool that allows organ–organ integration of hormonal signalling as a phenocopy of menstrual cycle and pregnancy-like endocrine loops and has great potential to be used in drug discovery and toxicology studies.
Non-invasive preimplantation genetic screening using array comparative genomic hybridization on spent culture media: a proof-of-concept pilot study
Overall, 17 out of 35 (48.6%) single chromosomal aneuploidies were concordant between the culture media and polar body analysis. By analysing negative controls (oocytes with fertilization failure), notable maternal contamination was observed. Therefore, non-invasive PGS could serve as a second matrix after polar body or cleavage stage PGS: however, in euploid results, maternal contamination needs to be considered and results interpreted with caut
Inositol and In Vitro Fertilization with Embryo Transfer
As shown by growing number of trials, MYO supplementation improves oocyte quality by reducing the number of degenerated and immature oocytes, in this way increasing the quality of embryos produced. Inositol can also improve the quality of sperm parameters in those patients affected by oligoasthenoteratozoospermia.
`Space pups` bred on Earth from mouse sperm stored on ISS for nine months
Freeze-dried sperm that was stored in space aboard the International Space Station (ISS) for nine months has been used to produce healthy mouse pups on Earth. The `space pup` experiment, which examined the effects of radiation on reproduction, marked the longest period mammalian sperm has been preserved in space.
3D-printed ovaries successfully restore mouse's fertility in holy grail of bioengineering
Scientists have used 3D-printed ovaries to successfully restore fertility for the first time in what they call the holy grail of bioengineering for regenerative medicine. A female mouse's ovaries were removed and replaced with 3D printed bioprosthetic ones using gelatin as the "ink" and using eggs from different mice — enabling it to ovulate, conceive pups and give birth, the United States researchers said.
