The relationship between gut and adipose hormones, and reproduction
Gut and adipose hormones interact with the reproductive axis as well as with each other. While leptin and insulin have stimulatory effects and ghrelin has inhibitory effects on hypothalamic GnRH secretion, there is increasing evidence for their roles in other sites of the reproductive axis as well as evidence for the roles of other gut and adipose hormones in the complex interplay between nutrition and reproduction. As our understanding improves,
Individualization of controlled ovarian stimulation in IVF using ovarian reserve markers: from theory to practice
Personalized IVF offers several benefits: it enables clinicians to give women more accurate information on their prognosis thus facilitating counselling especially in cases of extremes of ovarian response. The deployment of therapeutic strategies based on selective use of GnRH analogues and the fine tuning of the gonadotrophin dose on the basis of potential ovarian response in every single woman can allow for a safer and more effective IVF practi

Cadherin-dependent filopodia control preimplantation embryo compaction
Here, using live mouse embryo imaging, we demonstrate that cells extend long E-cadherin-dependent filopodia on to neighbouring cells, which control the cell shape changes necessary for compaction. We found that filopodia extension is tightly coordinated with cell elongation, whereas retraction occurs before cells become round again before dividing. Laser-based ablations revealed that filopodia are required to maintain elongated cell shapes. Moreo
Progesterone elevation and probability of pregnancy after IVF: a systematic review and meta-analysis of over 60 000 cycles
Based on the analysis of more than 60 000 cycles, it can be supported that PE on the day of hCG administration is associated with a decreased probability of pregnancy achievement in fresh IVF cycles in women undergoing ovarian stimulation using GnRH analogues and gonadotrophins. On the other hand, an adverse effect of PE does not seem to be present in frozen–thawed and donor/recipient cycles.
Can we prevent premature luteinization in IVF cycles?
From 1991 there is a long-lasting discussion on a possible detrimental effect of premature increase of progesterone levels during ovarian stimulation in IVF. A recent meta-analysis of more than 60,000 cycles states that premature progesterone increase reduces pregnancy rates starting from P4 values of 0.8ng/ml. The detrimental effect seems to be related to endometrial receptivity impairment according to lack of detrimental effect on oocytes compe
Association Between AMH, Oocyte Number and Availability of Embryos for Cryopreservation in IVF
Conclusion: AMH appears to be a valuable marker mainly for ovarian reserve and response to IVF treatment. AMH levels are strongly associated with the number of retrieved oocytes and the availability of supernumerary embryos suitable for cryopreservation.
Male contraception via simultaneous knockout of α:1A-adrenoceptors and P2X1-purinoceptors in mice
These results suggest that this autonomic method of male contraception appears free of major physiological and behavioral side effects. In addition, they provide conclusive proof of concept that pharmacological antagonism of the P2X1-purinoceptor and α:1A-adrenoceptor provides a safe and effective therapeutic target for a nonhormonal, readily reversible male contraceptive.
Out-of-Pocket Fertility Patient Expense: Data from a Multicenter Prospective Infertility Cohort
A total of 332 couples completed cost diaries and had data available on treatment and outcomes. Average age was 36.8 and 35.6 years in men and women, respectively. Of this cohort 19% received noncycle based therapy, 4% used ovulation induction medication only, 22% underwent intrauterine insemination and 55% underwent in vitro fertilization. The median overall out-of-pocket expense was $5,338 (IQR 1,197–19,840). Couples using medication only had t
Efficient generation of transgenic mice by lentivirus-mediated modification of spermatozoa
This relatively simple, yet highly efficient, technique for generating transgenic animals by transducing spermatozoa with lentiviral vectors in vitro is a powerful tool for the study of fertilization/preimplantation development, vertical viral gene transmission, gene function and regulation, and epigenetic inheritance.
IVF with planned single-embryo transfer versus IUI with ovarian stimulation in couples with unexplained subfertility: an economic analysis
. The mean cost per included couple was significantly different: €2781 with IVF–eSET and €1876 with IUI–ovarian stimulation (P < 0.01). The additional costs per ongoing pregnancy were €2456 for IVF–eSET. In couples with unexplained subfertility, one cycle of IVF–eSET cost an additional €900 per couple compared with three cycles of IUI–ovarian stimulation, for no increase in ongoing pregnancy rates or decrease in multiple pregnancies. When IVF–eSE
Strengths and pitfalls of Canadian gamete and embryo donor registries: searching for beneficent solutions
For the gamete and embryo donation community, it is well recognized that the implementation of a gamete and embryo donor registry (GEDR) represents a good initiative to ensure the best possible health conditions for donor-conceived individuals. Be they national, institutional or independent, GEDR can play a major role in the transmission of health-related genetic and medical information. However, from a bioethical analysis standpoint, GEDR raise
'Designer Sperm' Inserts Custom Genes Into Offspring
A new research report appearing online in The FASEB Journal, shows that introducing new genetic material via a viral vector into the sperm of mice leads to the presence and activity of those genes in the resulting embryos. This new genetic material is actually inherited, present and functioning through three generations of the mice tested. This discovery - if successful in humans - could lead to a new frontier in genetic medicine in which disease
Cell 'arms' reach out and pull early embryo into shape
'Now that we know what controls early development, we are designing non-invasive imaging approaches to see if human embryos used in IVF form normal filopodia and undergo normal compaction. This could help us choose which embryos should or shouldn't be implanted back in the uterus', Dr Plachta said.
Although the demand for infertility treatment is rising, the high cost may deter some couples from seeking care. Researchers from the University of California-San Francisco assessed direct out-of-pocket costs for couples undergoing fertility treatment. Those using medication only had the lowest out-of-pocket expenses at $912, while those using in vitro fertilization (IVF) had the highest at $19,234. The results, published in The Journal of Urolog
IVF Improving, but Fertility Treatments Keep Multiple Births High
Fertility technology in the United States has a huge influence on the frequency of twins, triplets, and other multiple births, according to new estimates published Dec. 5 in the New England Journal of Medicine. Dr. Eli Y. Adashi, professor of obstetrics and gynecology at Brown University, and his colleagues calculated that more than a third of twin births and more than three-quarters of triplets or higher-order births in the United States in 201
A University of Wisconsin-Madison research team reports today that the brain can produce and release estrogen - a discovery that may lead to a better understanding of hormonal changes observed from before birth throughout the entire aging process. The new research shows that the hypothalamus can directly control reproductive function in rhesus monkeys and very likely performs the same action in women.
Dual Protein Knockout Could Lead to New Male Contraceptive
A new male contraceptive could be on the horizon after scientists identified a novel way to block the transport of sperm during ejaculation. - Published today in the journal Proceedings of the National Academy of Science scientists have found that complete male infertility could be achieved by blocking two proteins found on the smooth muscle cells that trigger the transport of sperm.
